Hi,
Tell the folks at the store that you will be driving
for four to five hours and ask that they pack them accordingly.
If they do it right, they will decrease the nr of
fish/bag, and increase the bag size as well as amount
of water.
If they are bagged that way, they should, also, provide
the large styrofoam containers that the fish were
shipped to them in. When I worked at a LFS we used to
save them for just such occassions. Also we would
set them out in stacks for folks to purchase, singley,
for picnic coolers, or for occassions such as yours.

If you are going to take them in one container, free
swimming, like fish bait, then when at the store,
purchase a 12 volt air pump, tubing valves and
airstones. The device will plug into your
cigarette lighter socket and provide the air and
circulation within the containers.

Hi,
Well, unless you are going to keep them in your room
while on vacation, I'd drive to the LFS and then make
the long trip directly home to their new tank.
It's less stressful to them that way.

Hi,
Keep them under the least stress.
The absolute best for the fish is to go
with my second post. Drive all the way home
with them.
But...if you have to...
When you get to your room open the bags to the
air. In the morning add a glass of water, and
reseal them. Don't dump them into a tank, and
then pull them out to rebag them etc.
